Fermentation Services

In addition to our Lotus product offerings, Boston Mountain Biotech is a contract research organization (CRO) specializing in recombinant protein expression and other contract fermentation services. We are equipped to handle production in bacterial (ex. E. coli, bacillus, etc.), yeast, and insect cell systems in both batch fermentation and fed-batch fermentation setups. We can utilize an existing cell propagation protocol or we can work with you to design the protocol that best fits your needs. This can include designing and/or optimizing the media formulation, growth temperature, induction time, and expression vector.

The Fermentation Process

It is very important for the fermentation process to optimized in order for a bioreactor based system to provide optimal expression as well as being in a usable format. Improper fermentation optimization can cause the desired protein product to be truncated, improperly folded, insoluble, etc. These problems can exponentially increase the difficulty experienced during downstream protein purification. We can utilize an existing cell propagation protocol or we can work with you to design the protocol that best fits your needs. This can include designing and/or optimizing the media formulation, growth temperature, induction time, and expression vector.

Fed-batch fermentation protocols will utilize our 1-L, 3-L, or 7-L Applikon bioreactors which have full control systems (pH, temperature, dO2, stir rate, feed rate, antifoam, etc.).  Batch fermentation can be conducted in our Applikon bioreactors or in shake flasks (2L and smaller).

After fermentation has been completed, the next step is to proceed to protein purification. If you are not achieving desirable expression levels at this stage, one option is to re-evaluate your selected expression host or optimize your existing host.
